clp-1(RNAi) suppresses neurodegeneration of mec-4(d), deg-3(d), or gsa-1(gf) mutants; suppression by clp-1(RNAi) is enhanced by also applying tra-3(RNAi), but not by asp-3(RNAi) or asp-4(RNAi).
Moreover, neurodegeneration inflicted by cell-specific overexpression of asp-3 and asp-4 cannot be bypassed by a deficiency in either clp-1 or tra-3, implying that ASP-3 and ASP-4 are downstream of CLP-1 and TRA-3.
clp-1 is expressed strongly in many types of cell and tissue, including muscle cells and neurons.
One hypothesis to explain these data is that perturbation of intracellular concentrations of calcium may initiate neuronal degeneration by deregulating proteolysis.
