Phenotype it46ts mat : homozygous hermaphrodites produce 90% (16x) to 99% (25x) arrested embryos escapers sterile; abnormal early cleavages defective P granule localization. ES3 (progeny). NA4 (it5ts : weaker phenotype e2030 : grandchildless).

[C.elegansII] it46ts,mat : homozygous hermaphrodites produce 90% (16C) to 99% (25C) arrested embryos, escapers sterile; abnormal early cleavages, both spindles transverse at 2nd cleavage (failure of P1 rotation), defective P granule localization. ES3 (progeny). OA7: it5ts (weaker phenotype), e2030 (Mes, grandchildless),it87 (weak),jb2, etc. All ts; temp shift indicates not needed after 2nd cleavage. Cloned: encodes predicted novel protein. Antibody staining indicates specific asymmetric cortical localizations in early blastomeres. [Cheng et al. 1995; KK]

Structured_description Provisional_description par-2 encodes a protein containing a C3HC4-type RING-finger, a specialized type of zinc-finger found in E3 ubiquitin ligase subunits; PAR-2 is required maternally for polarization of the C. elegans zygote along the anterior-posterior (A-P) axis and is asymmetrically localized to the posterior cortex of the one- and two-cell embryo via interdependent interactions with PAR-3, PAR-6, and PKC-3. PAR-2 is also required for cortical contractions and cytoplasmic flow in the fertilized embryo, the asymmetric division of the zygote, known as P0, and the intrinsically programmed nuclear rotation of P lineage cells.
PAR-2 is thought to maintain A-P polarity in part by restricting the PAR-3/PAR-6/PKC-3 complex from the embryonic posterior after pronuclear meeting.
PAR-2 is also required for cortical and asymmetric localization of PAR-1 and enrichment of LET-99 in a cortical band in P lineage cells.
In addition to PAR-3, PAR-6, and PKC-3, PAR-2 localization is also dependent upon PAR-5 and NYM-2.
PAR-2 protein is excluded from the apical surface of embryonic cells by PAR-3 protein.
