adr-2(gv42) mutants show no ADAR activity either in vitro (from extracts of mixed-stage whole worms) on an artificial substrate, or in vivo on 256 sites of ADAR action on natural mRNA substrates.
Despite the complete loss of ADAR activity in adr-2(gv42) mutants, they display normal vulval development, in contrast to adr-1(gv6) mutants which show a mix of partial ADAR loss and partial ADAR hyperactivity.
The putative catalytic site of ADR-1 is divergent from the consensus sequence found in most ADARs (including ADR-2), which suggests that ADR-1 might form a catalytically inactive heteromeric subunit with ADR-2.
