The F-box motif consists of roughly 50 amino acids.
There are at least 11 F-box genes encoded by the genome of budding yeast, 22 in Drosophila, and 38 in human beings; but a striking 326 F-box genes are predicted to exist in C. elegans, comprising 1.6% of the protein-coding genome.
This disproportionate number of F-box genes is paralleled by the number of Skp-1 homologs (which are potential F-box ligands) encoded in these genomes: while there are 2, 7, and 1 Skp-1 related genes predicted in the genomes of Dictyostelium discoideum, Drosophila, and Homo sapiens, there are 17 Skp-1 homologs in C. elegans.
F-box proteins were first characterized as components of SCF ubiquitin-ligase complexes (named after their main components, Skp-1, Cullin, and an F-box protein), in which they bind substrates for ubiquitination (and, thus, subsequent proteolysis); in SCF complexes, the F-box motif links its protein to other SCF components by binding Skp-1 protein.
However, F-box proteins are by no means confined solely to SCF complexes: for instance, yeast Skp-1 can bind non-SCF F-box proteins to perform cellular functions completely distinct from protein degradation, such as kinetochore function (binding the F-box protein Ctf13p) or SNARE recycling (by binding the F-box protein Rcy1p), as well as yet other F-box proteins of unknown function (Yjl149 and Yml088).
