mdf-1 encodes a coiled-coil domain-containing protein that is orthologous to the Saccharomyces cerevisiae mitotic spindle checkpoint protein Mad1p; consistent with a role as a checkpoint protein, MDF-1 is required in C. elegans for mitotic genome stability and hence, long-term survival and fertility; in vitro, MDF-1 interacts with MDF-2, the C. elegans Mad2p ortholog, and with IFY-1, a Securin homolog; the maternal effect lethality produced by an mdf-1 mutation can be suppressed by a mutation in fzy-1, which encodes the C. elegans Cdc20 homolog. |
