cnx-1 encodes the C. elegans ortholog of calnexin, a transmembrane calcium-binding protein that localizes to the endoplasmic reticulum; by homology, CNX-1 is predicted to function as a molecular chaperone that is required for glycoprotein folding and maturation as well as for regulation of intracellular calcium homeostasis; although loss of cnx-1 activity via large-scale RNAi screens does not result in any obvious abnormalities, genetic studies indicate that cnx-1 RNAi can suppress the necrotic-like cell death induced by hyperactive mutations in the MEC-4 and DEG-1 ion channels; thus, in addition to its proposed role in glycoprotein processing, CNX-1 is also required for necrotic-like neuronal death. |
