The hcp-3 gene encodes a centromere protein (CENP)-A homolog required for kinetochore function; inactivation of hcp-3 in one-cell embryos by RNAi causes a complete loss of kinetochores, with total failure of chromosomes to segregate properly during mitosis, to recruit components to the kinetochore other than HCP-3, or to assemble a stable mitotic spindle; in addition, HCP-4 fails to localize properly to the kinetochore in hcp-3(RNAi) embryos. |
